A Study of Antifibrinolysin Activity

چکیده
This report describes methods which have been developed in this laboratory for the determination of antifibrinolysin activity in plasma. Using these methods, studies have been made of the antifibrinolysin activity in the plasma of the alligator (Alligator inississippiensis), guinea pig (genus Cavia), cow (Bos taurus), rat (Rattus norvegicus, Sprague-Dawley strain), frog (Rana pipiens), man (white, male), cat (Felis domestica, dog (Canis familiaris, mongrel breeds), rabbit (genus Lepus, mixed strains), opossum (Didelphis virginiana), chicken (genus Gallus, Plymouth Rock), and pigeon (Columbia livia). With the exception of man, both sexes were represented but pregnant animals were not utilized in the sampling. Graphs are presented which indicate the relationships between plasma concentration (antifibrinolysin units) and the time required for the clots to dissolve. With the help of these graphs it is possible to measure the unit antifibrinolysin activity in a plasma sample from any of the species which have been studied. The terminology used in this report will be the one recently suggested by Loomis, George and Ryder (1) and by Astrup and Permin (2). Considerable confusion has existed in the past regarding nomenclature and we believe that a uniform terminology will assist greatly in the development of this field. Antifibrinolysin (Fibrinolysin Inhibitor [3], serum antitryptase [4], antiplasmin [5], serum antiprotease [6] ) is present in the plasma of all species which we have tested. It is not yet available in a pure state and therefore its chemical characteristics are unknown. This substance or substances in plasma inactivate fibrinolysin. It is not certain whether antifibrinolysin forms an inactive complex with fibrinolysin or whether the in-

Fibrinolytic and antifibrinolytic activity in pregnancy.
Fibrinolytic activity and serum antifibrinolysin were estimated in normal pregnant women, during and after labour. The decreased fibrinolytic activity found during labour returned to non-pregnant levels within 24 hours of delivery. During the same period, the serum antifibrinolysin was rapidly diminished. It is suggested that the post-partum increase in fibrinolytic activity to non-pregnant lev...

Although the antifibrinolysin test as originally described (1) is only qualitative in nature (2), it has proved valuable in the study of infections caused by the f8-hemolytic streptococcus. However, the results obtained by different investigators have been difficult to interpret primarily because of failure to control the various factors participating in the fibrinolysin-antifibrinolysin reacti...

This report describes methods for the quantitative assay of human plasma profibrinolysin (plasminogen) and for the semi-quantitation of fibrinolytic factors in urine. The concentrations of plasma profibrinolysin, antifibrinolysin, fibrinogen and lytic factors in urine of patients with an active neoplasm and in pregnant women are compared with their concentrations in normal individuals. Although...

That haemolytic streptococci of human origin produce a substance capable of liquefying human fibrin was first demonstrated by Tillet and Garner in 1933. The fibrin from patients recently recovered from haemolytic streptococcal infection was in many cases found to be resistant to the action of this fibrinolysin. and confirmed these preliminary observations, and demonstrated antifibrinoly-sin in ...

Injection of ACTH or cortisone has no effect on the amount of fibrinolysin resulting from the activation of profibrinolysin by peptone. Antifibrinolytic activity of serum is significantly increased by ACTH or cortisone but not by desoxycorticosterone or corticosterone. No increase in anti-fibrinolytic power of serum is produced, however, by the injection of ACTH or cortisone in splenectomized g...
